摘要
目的:研究羟喜树碱对人肾上腺皮质癌SW-13细胞生长抑制和凋亡作用的影响。方法:采用CCK8检测不同浓度羟喜树碱对SW-13细胞增殖的影响;流式细胞仪测定药物作用前后的细胞凋亡情况。结果:不同浓度的羟喜树碱分别作用24,48,72h后,SW-13细胞的生长增殖均受到明显抑制(P<0.01),并呈时间和浓度依赖性;流式细胞术显示SW-13细胞凋亡率随羟喜树碱浓度的增加而升高。结论:羟喜树碱可以显著抑制人肾上腺皮质癌SW-13细胞生长,促进其凋亡。
Objective:To investigate the effect of hydroxycamptothecin (HCPT),a antineoplasticon,on the proliferation and apoptosis of human adrenal cortex carcinoma cell line SW-13.Methods:The cell-growth inhibition of HCPT on SW-13 cells was assessed by CCK8 assay.The cell apoptosis was measured by flow cytometry when treated with HCPT.Results:HCPT significantly inhibited the SW-13 cell proliferation (P〈0.01) with a time-and concentration-dependent manner.Flow cytometry displayed that the apoptosis rate of SW-13 cell increased with HCPT concentration increase.Conclusion:The results showed that HCPT not only can significantly inhibit the growth of adrenal cortical carcinoma SW 13 cells,but also promote apoptosis.
