靛蓝对溃疡性结肠炎体外炎症模型的作用 被引量:15

Study of indigo on ulcerative colitis inflammatory cell model in vitro

摘要 目的:研究青黛及锡类散主要成分靛蓝对溃疡性结肠炎(UC)体外炎症模型的抗炎作用。方法:以10μg/mL脂多糖(LPS)刺激RAW264.7细胞24h诱导UC体外炎症模型,以MTT法检测靛蓝的细胞毒性,以酶联免疫吸附法研究靛蓝在直接干预、预防干预2种模式下对细胞培养液IL-6/TNF-α表达的作用。结果:模型组中IL-6/TNF-α表达较正常组均有明显升高(P<0.05,P<0.01);靛蓝直接干预时,细胞培养液IL-6(5.0、10.0μmol/L实验组)/TNF-α(2.5μmol/L实验组)均较模型组有明显下降(P<0.05);靛蓝预防干预时,2.5μmol/L实验组中IL-6明显下降(P<0.05)。结论:靛蓝对UC有一定的体外抗炎作用,抗炎模式以直接干预为主,抗炎机制可能为下调IL-6的表达。 Objective: To investigate anti-inflammatory effect of indigo, which is main extractive of natural indigo and Xilei Powder, on inflammatory cell model of ulcerative colitis (UC). Methods: Murine macrophage RAW264.7 cells were induced by 10lag/mL LPS for 24h to establish an UC inflammatory cell model. Cell viability was detected by MTT assay to explore the safe range of concentrations. UC inflammatory cell model was treated by indigo in two different ways: treatment mode and prevention mode. IL-6 and TNF-α in supernatant was tested by Elisa assay. Results: IL-6 and TNF-α in supernatant in model group were significantly higher than control group (P〈0.05, P〈0.01); Under treatment model, IL-6 and TNF-a in supernatant were reduced by indigo significantly (P〈0.05); While under prevention mode, only IL-6 was reduced (P〈0.05). Conclusion: Indigo could inhibit inflammation in UC inflammatory cell model in vitro, mainly under treatment mode by down-regulating IL-6 expression.
