响应面法优化稻壳中多糖的微波辅助酶提取工艺

摘要 为优化稻壳多糖的微波辅助酶法提取工艺,在单因素实验的基础上,选择纤维素酶添加量、微波功率及料液比3个因素的Box—Behnken中心组合实验设计,以多糖得率为响应值,采用响应面分析法优化稻壳多糖的提取工艺,模拟得到二次多项式回归方程的预测模型。结果表明:稻壳多糖提取的最佳工艺为纤维素酶添加量1.00%,料液比1:29(g:mL),微波功率560W,微波时间2min,在此条件下稻壳多糖的实际得率达1.127%。 In order to optimize the extraction method of polysaccharide from rice hull by microware-assisted enzyme extraction technology, on the basis of single- factor experiments, cellulose addition amount, microwave powder and solid-solvent ratio were selected as influencing factors and the content of polysaccharides as response during extraction.The experiment method was designed according to Box-Behnken central composite experiment, which studied each variable and their interactions effects on the contents of polysaccharide. The quadratic multinomial model on the contents of polysaccharide was established by using SAS software. The optimum condition of extracting process was determined by means of response surface method. The results showed that the optimum extraction conditions for polysaccharide were cellulose addition amount 1.00%, solid- solvent ratio ] : 29 ( g : mL), microwave powder 560W and microwave time 2min.Under the optimal extraction conditions,the maximum yield of polysaccharide was 1.127%.
