摘要
流式细胞术是20世纪70年代发展起来的一种快速的细胞定量分析和分选的新技术,具有测量速度快、高灵敏度、高精确度、多参数分析、高纯度分选的特点。过敏性疾病为威胁人类生命健康的疾病之一,但其诊断通常依赖病史以及适当的确证试验,但这些确证实验尚缺乏有力确证依据。过敏反应发生时,嗜碱性粒细胞表面分子可发生相应变化,通过对其表面标志物的检测,可达到对过敏性疾病进行鉴别诊断的目的。本文就流式细胞术分析嗜碱性粒细胞活化试验的几种设门筛选嗜碱性粒细胞思路加以介绍,并对流式细胞术在过敏性疾病诊断中的应用价值做一综述,从而为流式细胞术在过敏性疾病诊断领域中广阔的应用前景提供一有力的理论支持。
Flow cytometry is a new technology for rapid quantitative analysis and sorting,which is developed during 1970s.It has many advantages such as rapid speed,sensitivity,accuracy,multi parameters,and high purity selection.Allergic disease is one of the diseases which is threatening human life and health.Allergic disease diagnosis usually depends on the history and the confirmatory test.However,those tests are lack of proven evidence.When allergic reaction occurs,basophils surface molecules will change correspondingly,so that detect these surface markers can be a feasible method of differential diagnosis of allergic disease.This paper introduces several gating strategies in a flow cytometry analyzing basophil activation test,and also reviews of the application value,in order to provide more powerful theoretical support for flow cytometry in the diagnosis of allergic disease.
