摘要
目的:研制1种快速检测沙门氏菌、大肠杆菌O157∶H7和金黄色葡萄球菌的三重PCR检测试剂盒,并加以应用。方法:设计3种待检目的菌特异性引物,优化PCR反应条件,建立多重PCR检测试剂盒,并对其特异性、灵敏度、抗干扰性、准确性等方面进行评估。结果:该试剂盒特异性好,灵敏度高,3种待检目的菌在不同食品介质中的检出限均达到101CFU/mL,抗干扰性强。在6个月保存期内,其稳定性和准确性均未发生改变。结论:本研究建立的试剂盒可同时检测上述3种病原菌,且具有稳定性好、准确性高等特点,可用于禽肉及其制品的日常微生物安全监测。
Objective: A kit for simultaneously detection of Salmonella enteritidis, Escherichia coli O157:H7, Staphy-lococcus aureus by multiplex PCR was developed and applied. Methods: Through designing specific primers of three tar-get bacterias and optimizing PCR reaction conditions, multiplex PCR diagnostic kit was developed and subsequently eval-uated the specificity, sensitivity, anti-jamming capability and accuracy of this method. Results: The established kit was specific and highly sensitive. The detection limits of the three target bacterias were all as low as 10^1 CFU/mL in different food matrixes. The anti-jamming capability of the kit was also strong and the stability and accuracy of it were not changed in six months. Conclusion: The developed kit in this study enables simultaneous detection of three pathogens described above and has the characteristics of stability, accuracy and others. It can be used to monitor daily microbial safety of poultry and their products.
