Expression of heparanase and its biological significance in renal cell carcinoma

摘要 目的:通过检测乙酰肝素酶(Heparanase,Hpa)在肾癌中的表达,探讨其生物学意义。方法:采用免疫组化PV6000法检测54例肾癌和20例正常肾脏组织中Hpa蛋白的表达,分析Hpa与肾癌临床病理特征的关系。结果:Hpa在肾癌组织中的表达较正常肾脏组织明显增高(P<0.05)。Hpa的表达水平与肾癌的临床分期、肿瘤大小呈正相关(P<0.05),而与肿瘤的组织学分级无相关性(P>0.05)。有转移的肾癌患者中Hpa的阳性表达率高于无转移者(P<0.05)。Hpa的表达与患者年龄、性别、病理学类型无相关性(P>0.05)。结论:Hpa与肾癌的发病及浸润转移等不良预后相关,并有可能成为判断肾癌预后的指标和靶向治疗的靶点。 Objective To explore the expression of heparanase in human renal cell carcinoma and its correlation with clinical fea- tures. Method The expressions of heparanase in 54 renal cell carcinoma and 20 normal renal tissues were assessed by PV6000 of immuno- histochemistry to do statistical analysis for the correlation between their clinicopathologic features of those clinical cases. Results The ex- pression of heparanase protein significantly increased in renal cell carcinoma compared with that in normal renal tissues(P 〈 0. 05). The ex- pression of heparanase protein was positively correlated to the clinical stages and tumor diameter of RCC (P 〈 0. 05), but no correlation be- tween its expression and the pathological grades of renal cell carcinoma( P 〉 0. 05 ). The positive expression rate of heparanase in patients with distant metastasis was higher than that in patients without distant metastasis,there was significant difference(P 〈 0. 05). There were no associations between the expression of heparanase protein and ages, sexes of patients ( P 〉 0. 05). Conclusion The expression of heparanase protein significantly increases in renal cell carcinoma compared with that in normal renal tissues. It suggests that they may play an important role in renal carcinogenesis and progression.
