摘要
目的:测定纳米银冻干粉小鼠经口给药的最大耐受剂量。方法:按照最大耐受剂量测定法,将ICR小鼠40只随机分为对照组和实验组,每组20只,实验组采用最大浓度、最大容积的纳米银冻干粉一次经口给药,对照组给予等量生理盐水,观察小鼠给药后2周内的行为、外观、进食、排泄物、主要脏器的大体和病理学改变以及死亡情况,并计算小鼠对纳米银的最大耐受量。结果:纳米银冻干粉小鼠经口染毒后,行为无异常,摄食量、体重等与对照组比较差异无统计学意义(P>0.05),各主要脏器无明显异常改变,2周内无死亡。结论:采用最大浓度、最大容积的纳米银冻干粉行小鼠经口给药最大耐受量试验,经折算最大耐受剂量为临床成人日用量的3 592倍。
Objective: To determinate the maximum tolerated dose (MTD) of nano silver lyophilized powder administered orally in mice. Methods: According to the determination of the MTD, 40 ICR mice were randomly divided into experimental group and control group. The experimental group was treated with nano silver lyophilized powder with gastric infusion at maximum concentration and volume, while control group was given equal dose of normal saline. The behavior, appearance, food intake, excrement, the general and pathological changes of major organs and death were observed in both groups within two weeks after gastric infusion, and the MTD in mice was converted. Results: After administering of nano silver lyophilized powder, no obvious abnormality behavior was found in all mice. Food intake and body weight in experimental group showed no significant difference compared with the control group(P 〉0. 05). No deaths within 2 weeks in both groups. Condusion: The MDT is as 3 592 times as clinical human adult daily dose after converting based on the orally administering of nano silver lyophilized powder with maximum concentration and volume in mice.
