摘要
目的了解丰台区流行的麻疹野病毒的基因特征,为控制和消除麻疹提供依据。方法采集2013年春季疑似麻疹爆发和散发患者咽拭子标本分离病毒,通过逆转录一聚合酶链反应(RT-PCR),从分离到的麻疹病毒中扩增出核蛋白(nucleoprotein,N)基因C末端634个核苷酸片段,再对扩增产物进行核苷酸序列测定和分析,并以C末端450个核苷酸片段构建基因亲缘关系树,进行遗传距离分析。结果共分离麻疹病毒39株,测序后为Hl基因型(28株)和D8基因型(11株)。结论丰台区2013年春季麻疹流行株仍属于H基因组,Hl基因型,Hla亚型。此外北京市丰台区首次发现D8基因型麻疹野病毒。
Objective To investigate genotype of wild-type measles viruses circulated in Fengtai district in the spring of 2013, and provide basis for measles control and elimination. Methods Measles viruses were isolated from the specimens collected from suspected measles outbreaks and sporadic patients. Fragments of 634 nucleotide acids of the carboxyl end of nucleoprotein gene were amplified using reverse transcription--polymerase chain reaction (RT--PCR) method, and thereafter, the PCR products were directly sequenced and analyzed. Finally the phylogenetic tree was constructed based on 450 nucleotide acids of the carboxyl end of nucleoprotein gene, and the diversity analysis was done with MEGA version. Results A total of 39 wild-type measles viruses were isolated and sequenced. Among which,2$ strains were identified as H 1 genotype, and 11 strains were identified as D8 genotype. Conclusions Major genotypes of wild-type measles viruses circulated in Fengtai district, Beijing in 2013 were Hla genotype. In addition, D8 genotype measles virus was found for the first time in Fengtai district,Beijing.
