摘要
免疫PCR方法是结合免疫学和PCR原理的一种新型检测技术,具有高灵敏度、高自动化、操作简便等特点。研究采用PCR方法建立饮料中赤藓红快速检测方法。通过优化包被抗原浓度、探针DNA的量、抗体浓度等条件,结果显示:赤藓红的检测低限为10.0fg/g,线性浓度范围为0.05~50pg/g,果汁饮料中赤藓红的添加回收率为71.5%~112.6%,该方法简单、灵敏,适用于饮料中赤藓红的测定。
Immuno-polymerase chain reaction(Immuno-PCR) is a novel technique with the characteristic of high sensitivity,high automa tion and easy operation. A rapid detection method of erythrosine was developed. After the optimization of the coating antigen concentra- tion, the amounts of probe DNA, and the antibody concentration, erythrosine detection limit was low as 10.0 fg/g, and the linear con centration range was 0.05 - 50 pg/g. The recoveries of fruit drinks ranged from 71.5% to 112.6%. This method was simple, sensitive, and suitable for the detection of erythrosine in drinks.
