摘要
目的研究地锦草对H22荷瘤小鼠的抑瘤作用并探讨其VEGF、MMP-9蛋白表达的影响。方法建立H22肝癌皮下移植瘤模型,随机分为模型对照组、地锦草高剂量组(265 mg/kg)、地锦草中剂量组(132mg/kg)、地锦草低剂量组(66 mg/kg)、环磷酰胺组(50 mg/kg)。观察小鼠的般状态和肿瘤生长情况,灌胃给药14 d后处死小鼠,分别剥离瘤组织,测量肿瘤体积大小。免疫组化法观察各组肿瘤组织中VEGF、MMP-9蛋白表达情况。结果地锦草治疗组小鼠般情况良好,肿瘤体积明显缩小(P<0.05);免疫组化结果显示:VEGF、MMP-9阳性表达定位于细胞浆中,呈棕黄、棕褐及浅黄色,胞质染色。模型组VEGF、MMP-9蛋白表达增强,地锦草与环磷酰胺组VEGF、MMP-9蛋白表达下降(P<0.05);且VEGF、MMP-9蛋白表达无直接相关性。结论地锦草能抑制荷瘤小鼠的肿瘤的生长。抑制荷瘤小鼠肿瘤组织的VEGF、MMP-9蛋白的表达,从而减少肿瘤的血管生成和基质的降解可能是其部分机制之。
[ Objective ] To study the effects of Euphorbiae humifusae herb(EHH) on antitumor and the expression of vascular endothelial growth factor (VEGF), matrix metalloproteinases-9 (MMP-9) on H22 cells beating mice. [Methods] H22 liver cancer cells were inoculated in the right axilla to prepare solid tumor models. Five groups were randomized in each batch, named model control group, a high-dose group, a middle dose group and a low-dose group, a positive control group. Different doses of EHH (265, 132, 66 mg/kg) were ip administered to H22 cells beating mice once a day for continuous 14 d. The tumor was weighted, while histopathological morphology of the tu- mor was observed under optical microscope. Using immunohistoehemieal method, the protein expression of VEGF and MMP-9 in tumor tissues was determined. [ Results ] The size of tumor in EHH treat groups was much smaller than that in control group (P 〈0.05). The immunohistochemical results showed that the expressions of VEGF and MMP-9 were decreased in EHH treat groups (P 〈0.05). [Conclusion] EHH acts on the inhibition of H22 cells bearing mice and its mechanism is probably related to decrease the expressions of VEGF and MMP-9 in H22 cells bearing mice.
