ROCK抑制剂对人食管癌EC9706细胞迁移能力的影响 被引量:4

Effect of ROCK inhibitor on the migration ability of human esophageal cancer EC9706 cells in vitro

摘要 目的研究ROCK特异性抑制剂Y-27632对人食管癌EC9706细胞迁移能力的影响,并探讨其可能的机制。方法体外培养人食管癌株EC9706,加入20μmol/L的Y-27632溶液(实验组)及等体积的PBS(对照组)培养1 h,采用细胞计数、MTT实验检测细胞生长情况,采用Transwell实验观察细胞迁移情况,采用Western blot检测Cav-1的表达水平,并进行两组的比较。结果细胞计数、MTT、Transwell实验结果显示,Y-27632可以抑制细胞的生长与迁移能力,与对照组比较差异有统计学意义(P均<0.05)。Western blot检测结果显示,Y-27632处理细胞1 h后,Cav-1蛋白表达水平明显下降,并明显低于对照组(P均<0.05)。结论 ROCK信号通路参与EC9706细胞生长与迁移的调节,Y-27632可以抑制EC9706细胞的生长及迁移能力,其机制可能与下调Cav-1的表达有关。 Objective To investigate the effect of Y-27632, a specific inhibitor of Rho-associated coiled- coil forming protein kinase (ROCK) ,on migration ability of human esophageal cancer EC9706 cells and explore its potential mechanism. Methods Twenty μ mol/L Y-27632 solution ( medication group) and equal volume PBS solu- tion ( control group) were added in the EC9706 cells cultured in vitro, respectively, and cultured for 1 h. The growth of the cells was detected by cell counting and MTY method ; the cell migration was observed by Transwell test; the Caveolin-1 (Cav-1) expression level was detected by Western blot method. The aforementioned results were com- pared in two groups. Results The results of cell counting, MTT and Transwell tests showed that Y-27632 could in- hibit the proliferation and migration of cells compared with control group ( all P 〈 0.05 ), and Western blot test showed that after the cells were treated with Y-27632 for I h, the expression level of Car-1 significantly decreased to the level less than control group ( P 〈 0.05 ). Conclusions ROCK signaling pathway might take part in the regula- tion of growth and migration ability. Y-27632 can inhibit the growth and migration ability of EC9706 cells, and its mechanism may be related to down-regulation of Cav-1 expression.
