摘要
目的:探讨中药方剂肾衰泻浊丸对UUO大鼠模型肾组织CTGF和α-SMA表达水平的影响,初步探讨其对肾间质纤维化的作用机制。方法:采用单侧输尿管梗阻致肾间质纤维化大鼠模型。分为假手术组、模型组、肾衰泻浊丸高剂量组、肾衰泻浊丸中剂量组、肾衰泻浊丸低剂量组、洛汀新组,运用免疫组化法检测实验第14、21、28天各组的CTGF和α-SMA的表达水平。结果:不同剂量的肾衰泻浊丸组和洛汀新组可明显下调α-SMA和CTGF在实验大鼠肾小管、间质的表达,与模型组相比有显著性差异(P<0.05),且中剂量组效果最优,与洛汀新组相比有显著性差异(P<0.05)。结论:肾衰泻浊丸可以降低实验大鼠肾组织中α-SMA和CTGF的表达水平,延缓肾间质纤维化的进展。
Objective :To investigate the effect of the Shenshuai Xiezhuo Pill on CTGF and α - SMA of kidney tissue of renal interstitial fibrosis rats caused by unilateral urethral obstruction ( UUO), and to illustrate the mechanism of Shens- huai Xiezhuo Pill on delaying renal interstitial fibrosis. Methods : The rats were divided into six groups, including sham - operated group, model control, high, middle and low doses groups of Shenshuai Xiezhuo Pill, and Benazepril group. CTGF andα - SMA expressions in kidney tissues were detected by immunohistochemical method on 14,21,28 days. Results : Shenshuai Xiezhuo pill can significantly lower expressions of CTGF and a - SMA in tubulointerstitial and had obvious difference with control group. Shenshuai Xiezhuo pill can improve the renal tubulointerstitial pathological lesions. Conclu- s/ons : Shenshuai Xiezhuo pill can significantly lower expressions of CTGF and α- SMA in tubulointerstitial of the unilater- al urethral obstruction animal model, and delay the progression of the renal interstitial fibrosis.
