探讨野百合碱诱导大鼠右心室心肌肥厚模型的方法 被引量:1

摘要 目的探讨以野百合碱(Monocrotaline,MCT)诱导大鼠右心室心肌肥厚模型的制备方法。方法将SD雄性大鼠随机分为对照组(CON组)和野百合碱给药模型组(MCT组),MCT组以2%MCT按60mg·kg-1剂量一次性腹腔注射后饲养3周建立MCT诱导的右心室肥厚大鼠模型。分别测定CON组和MCT组的以下指标:①通过右心室插管法测定平均右心室内压(mRVP)、右心室内压力最大上升速率(+dp/dtmax)、右心室内压力最大下降速率(-dp/dtmax);②测定右心肥厚指数(RVMI);③HE染色观察右心室心肌组织切片。结果与CON组相比:①MCT组的mRVP及RVMI从显著升高(P<0.01,n=18);②右心室+dp/dtmax显著升高(P<0.01,n=18),右心室-dp/dtmax显著降低(P<0.01,n=18);③MCT组右心室心肌纤维粗大,细胞内肌原纤维数量增多,心肌纤维排列紊乱,细胞核深染,形状不整。结论 MCT诱导3周可成功诱导SD大鼠产生右心室心肌肥厚。 OBJECTIVE To explore the approach of right ventricular hypertrophy induced by Monocrotaline. METHODS The SD rats were randomly divided into control group (CON) and Monocrotaline administration (MCT group). MCT group, a dose of 50mg . kg-1 IN-MCT was given to SD rats by single intraperitoneaI injection and raised 3 weeks. In both models, We measured: ①Mean Right ventricular systolic pressure ( mRVP), right ventricular pressure maximum rate of rise ( RV + dp/dt,,. ), right ventricular pressure maximum rate of descent ( RV-dp/ dtmax)Right ventricular mass index (RVMI) ;③Right ventricular myocardium tissue sections were stained by HE and observed under light microscope. RESULTS In comparison to the CON group:(,~)mRYP and RVMI were markedly elevated (P 〈 0. 01 ) in MCT group ; ②RV + dp/dt.,, were markedly elevated ( P 〈 0. 01 ), RV-dp/dt.,= were markedly decreased (P 〈 0. 01 ) in MCT group ;(~Right ventricular myocardial fibers become thick, intracellu- lar myofibrils increase in the number of disordered arrangement of the myocardial fibers, the nuclei deeply stained, the shape of the nuclei was not regularity in MCT groups. CONCLUSION 3 weeks injected MCT could induce SD rats suffered right ventricular hypertrophy successfully.
