摘要
目的探讨流式细胞术(FCM)在蜀葵花抑制金黄色葡萄球菌(金葡菌)药敏试验中的应用价值。方法应用FCM检测蜀葵花对金葡菌的抗菌活性,药物作用3 h后,用碘化丙啶(PI)染色,分别检测10株金葡菌阴性对照管和最低抑菌浓度(MIC)药物浓度作用管的平均荧光强度(MFI),计算两者比值并设定流式细胞药敏试验(FCST)检测MIC的判断值,根据该判断值对21株临床菌株同时进行常规药敏和FCST双盲测定。结果 10株金葡菌测得的MFI比值为2.95±0.117,将MFI大于阴性对照管3倍(增加200%)的最低药物浓度定义为该菌的MIC;21株临床菌株中20株两种方法测得MIC值相差在1个倍比稀释度以内,两者比较无显著性差异(P>0.05)。结论 FCST与常规药敏试验结果有很好的一致性,并具有快速、敏感、准确的优势。
Objective It is to observe the application value of Flow Cytometry (FCM) in detection of the antibiotic suscep- tibility testing for the bacteriostasis of Hollyhock Flower to staphylococcus aureus. Methods The bacteriostasis of Hollyhock Flower to staphylococcus aureus was detected by FCM. After treated with homologue drug for 3 hours then they were stained by propidium iodide ( PI). The mean fluorescence intensity (MFI) of I0 staphylococcus aureus negative control and reaction tube for drug minimal inhibitory concentration (MIC) was detected respectively. According to the judgement level of MIC by Flow cytometry susceptibility testing (FCST) , 21 clinical isolates were double -blind examined at the same time by M27 -A meth- od and FCST. Results The ratios for MFI of I0 Staphylococcus aureus isolates was the MIC was defined as the lowest concentration of drug that showed an increase of 200% in MFI compared to that of the negative control. Comparing the MICs of FCST and M27 -A method in 21 clinical isolates, the difference of MIC value for 20 isolates was within I dilution, there was no significant difference in MIC between FCST and M27 - A method (P 〉 0.05 ). Conclusion FCST and M27 - A method are in excellent agreement, and FCST is a rapid accurate, sensitive and convenient method.
