摘要
目的考察整肠生菌株在传代过程中的遗传稳定性,研究其耐药性与质粒的关系。方法联合使用表型和RAPD测试,对整肠生菌株在传代过程中的遗传稳定性进行分析,对菌株进行抗生素药敏试验和质粒检测,利用溴化乙锭消除质粒,比较消除前后耐药性的改变。结果在传代过程中不同代次的整肠生菌株表型特征和遗传学特性无变化,整肠生菌株无质粒。结论整肠生菌株在25代以内遗传稳定,能安全用于生产,菌株耐药性与质粒无直接相关性。
Objective To analyse the genetic stability of Bacillus Licheniformis from ZhengCbangSheng(ZCS), and study the relation between its antibiotic resistance and plasmids. Methods The change of the different samples from different period during subculture were evaluated using phenotypic and RAPD tests. Antibiotic susceptibility of Bacillus Licheniformis was tested, the plasmids of strains were examined and to make a comparison of antibiotics susceptibility before and after elimination of plasmids. Results During subcuhure, Bacillus Licheniformis from dif- ferent period hold a good phenotypic and genetic stability and do not carry plasmids. Conclusion Bacillus Lichen# formis from ZCS can completely fulfil the demands of production in 25 generations with a high stability of the genetic traits. The antibiotic resistance of Bacillus Licheniformis is not mediated by plasmids.
