摘要
研究人源肠内菌转化"通脉方"的化学成分。采用硅胶、Sephadex LH-20、半制备性高效液相等柱色谱和重结晶等方法进行分离纯化,根据MS和NMR等谱学数据鉴定化合物的结构。分离鉴定了26个化合物,分别为苯丙酸(1)、6″-O-乙酰基葛根素(2)、芒柄花素(3)、大豆苷元(4)、对羟基苯丙酸(5)、吲哚-3-丙酸(6)、染料木素(7)、异芒柄花素(8)、异芒柄花苷(9)、(-)-野葛醇B-2″-O-吡喃葡萄糖苷(10a)和(+)-野葛醇B-2″-O-吡喃葡萄糖苷(10b)混合物、8-羟基大豆苷元(11)、野葛醇A(12)、3'-甲氧基-6″-O-乙酰基葛根素(13)、6″-O-乙酰基大豆苷(14)、3'-甲氧基大豆苷(15)、野葛醇B(16)、3-甲基尿嘧啶(17)、染料木苷(18)、大豆苷(19)、3'-甲氧基葛根素(20)、葛根素芹菜糖苷(21)、獐芽菜苦苷(22)、大豆苷元-7,4'-O-双葡萄糖苷(23)、腺嘌呤(24)、3'-羟基葛根素(25)和葛根素(26)。"通脉方"经人源肠内菌生物转化,所含苷类化合物被转化为苷元和/或少糖基化合物,亦包括羟基化反应和脱甲基反应;苷类化合物是"前药"。
To study the chemical constituents in Tongmai formula (TMF) after biotransformation by human intestinal flora (HIF) , water extract of TMF was anaerobically incubated with HIF at 37 ℃. Column chromatographic methods over silica gel, Sepha- dex LH-20 and semi-preparative high-performance liquid chromatography as well as recrystallization were used to isolate and purify the chemical constituents in TMF after biotransformation by HIF. The chemical structures of isolated compounds were identified on the ba- sis of MS and NMR data. Twenty-six compounds were obtained and identified as phenylpropionic acid(l), 6"-O-acetylpuerarin(2), formononetin ( 3 ), daidzein ( 4 ), p-hydroxyphenylpropionic acid ( 5 ), 3-indolepropionic acid ( 6 ), genistein ( 7 ), isoformononetin ( 8 ), isoononin(9), a mixture of ( - )-puerol B-2"-O-glucopyranoside(10a) and ( + )-puerol B-2"-O-glucopyranoside(10b), 8-hydroxy- daidzein( 11 ), puerol A ( 12 ), 3 '-methoxy-6"-O-acetylpuerarin ( 13 ), 6"-O-acetyldaidzin ( 14 ), 3 '-methoxydaidzin ( 15 ), puerol B ( 16 ), 3-methyluracil( 17 ), genistin ( 18 ), daidzin ( 19 ), 3 '-methoxypuerarin ( 20 ), mirificin ( 21 ), swertiamarin ( 22 ), daidzein-7, 4'-O-glucoside ( 23 ), adenine ( 24 ), 3 '-hydroxypuerarin ( 25 ), and puerarin ( 26 ). After biotransformation by HIF, the glycosides in TMF were transformed into aglycone and/or less glycosyl compounds along with some hydroxylation and demethylation reactions. There- fore, the glycosides in the TMF are the pro-drug.
