摘要
扫描电子显微镜(SEM)具有观察生物三维结构图象的能力,但对深部结构及细胞细胞器等的显示有困难。本实验应用多种特殊制片技术,可在SEM制片上清楚地显示下列结构:微血管三维结构、运动终板;平滑肌和弹力纤维在器官内的三维构筑以及基底膜等。这大大扩大了SEM在生物医学研究上的应用范围。
SEM has a capacity to observe biological surfaces in three—dimensions, with a great depth of focus of high resolution,whereas SEM has been largely limited to the observation of the hidden surfaces of the tissues and organes as well as the intracellular organelles. Therefore, this study presents several special preparative methods for SEM; methods to disclose intracellalar structures, corrosion casting methods to study architecture of blood vessels,motor end—plate,three-dimensions structure and arrangement of smooth muscles, elastic and collagen fibres and basal lamina. These techniques extend the application of the SEM in the biomedical field.
