摘要
目的建立能同时检测克宁松乳膏中氯霉素和地塞米松磷酸钠含量的方法。方法采用高效液相色谱法,以十八烷基硅烷键合硅胶柱(Diamonsil C18柱,5μm,4.6 mm×200 mm)为色谱柱;0.2%三乙胺水溶液(用磷酸调节pH至2.8)-乙腈(70∶30)为流动相;流速1.0 mL/min;检测波长256 nm。结果氯霉素和地塞米松磷酸钠的含量分别在24.577～786.448μg/mL(r2=0.999 9)和5.101 6～163.251 2μg/mL(r2=0.999 9)的范围内,与各自峰面积积分值呈良好的线性关系;平均回收率均为100%,RSD分别为1.2和1%(n=6)。结论用高效液相色谱法同时测定克宁松乳膏剂中的氯霉素和地塞米松磷酸钠含量,方法快捷、结果准确,而且稳定性好,不受辅料中基质的干扰。
Objective To simultaneously determine the contents of chloramphenicol and dexamethasone sodium phosphate in Keningsong cream with an HPLC method.Methods It was assayed by HPLC.The column was Diamonsil C18,5 μm,4.6 mm×200 mm;the mobile phase was buffer solution of TEA（use phosphoric acid adjust pH to 2.8）-acetonitrile（70∶30）,the flow rate was 1.0 mL/min,the UV detection wavelength was 256 nm.Results The linear range of Chloramphenicol was 24.577-786.448 μg/mL（r^2=0.999 9）,and Dexamethasone Sodium Phosphate was 5.101 6~163.251 2 μg/mL（r^2=0.999 9）.The average recoveries both were 100%,RSDs were 1.2 and 1%（n=6）,respectively.Conclusion Simultaneous determination of the content of chloramphenicol and dexamethasone sodium phosphate in Keningsong cream.The results obtained showed that it was a reliable and accurate method for the quality control of Keningsong cream.This method can get the content of Chloramphenicol and in Keningsong cream by HPLC in the same time,and free from the interference of the excipients matrix.
