摘要
目的:探讨健脾增液汤对2型糖尿病大鼠脂肪-胰岛素轴的早期干预作用。方法 :大鼠随机抽以10只为正常组,其余大鼠腹腔注射小剂量链脲佐菌素加高糖高脂饲料建立2型糖尿病大鼠模型,成模后,将模型动物随机分为模型组、二甲双胍组(0.25mg/kg)、健脾增液汤组(18g/kg)、健脾化湿组(9g/kg)、增液汤组(9g/kg),治疗8周,应用放射免疫法检测在大鼠血清瘦素(Leptin)水平、胰岛素(INS)水平,生化方法检测血脂、血游离脂肪酸(FFA)的含量,分析血脂、Leptin、INS在糖尿病发病中的相互关系,并以二甲双胍作为对照,分析了中药健脾增液汤及其拆方对糖尿病脂肪-胰岛素轴的早期干预作用。结果 :(1)各组大鼠血脂中总胆固醇(TC)、甘油三酯(TG)、FFA升高,高密度脂蛋白降低,瘦素水平、胰岛素水平均升高,与正常组有显著性差异。(2)健脾增液汤组(18g/kg)、健脾化湿组(9g/kg)、增液汤组(9g/kg)的高密度脂蛋白(HDL)升高,FFA、胰岛素、瘦素降低,而健脾增液汤组(18g/kg)、健脾化湿组(9g/kg)组的为TG、TC降低显著。结论:各组药物均可改善糖尿病的脂代谢异常,改善脂肪-胰岛素轴的功能,从而对因脂毒性、胰岛素抵抗引起的糖尿病的发生和发展有一定的干预作用。
Objective: To observe the effects of Janpizengye soup on fat-insulin-axis in the obese-rats of type 2 diabetes.Methods: The model of type 2 diabetes was established by intraperitoneal injection of streptozotocin(STZ) at a dose of 30mg / kg and a high-carbohydrate / high-fat diet.Experimental rats were randomly divided into six groups: normal group,model group,metformin group,combination group,invigorating spleen to resolve dampness group,Zengye soup group and they were treated respectively.Eight weeks later,Leptin 、INS、FFA、Lipids contents were determined through radioimmunoassay and Biochemical method respectively.Results :(1) Total cholesterol(TC),triglyceride(TG),FFA and leptin levels、insulin levels were elevated,there is a significant difference with the normal group(P < 0.05)(2) After treatment,FFA、insulin、leptin were decreased(P < 0.05) in combination group(18g / kg),invigorating spleen to resolve dampness group(9g/kg),Zengye soup group(9g/kg),TG、TC were also decreased in combination group(18g/kg),invigorating spleen to resolve dampness group(9g / kg).Conclusion: Invigorating spleen and zengye soup can improve fat-insulin-axis function,which due to improve lipid toxicity,insulin resistance,leptin resistance.
