黄芪甲苷对心肌缺血心律失常大鼠模型Ca^(2+)浓度影响初步研究 被引量:18

The Influence of Astragaloside Ⅳ on Myocardial Ischemia and Arrhythmia in Rat Model Myocardial Ca^(2+) Concentration and Its Anti-arrhythmia Mechanism

摘要 目的:初步探讨黄芪甲苷对心肌缺血心律失常模型大鼠心肌细胞Ca2+浓度的影响。方法:将心肌缺血再灌注模型大鼠随机分为模型组和假手术组,各组给药7d后开始造模。将Fura-2/AM荧光探针负载于大鼠心肌细胞,采用荧光分光光度计检测Ca2+浓度。结果:模型组心肌细胞内游离Ca2+浓度与假手术组比较,差异有显著性(P<0.01);与黄芪甲苷的低剂量组比较,无显著性差异(P>0.05);与黄芪甲苷的中、高剂量组比较,有显著性差异(P<0.01);黄芪甲苷的中、高剂量组与普罗帕酮组比较,无显著性差异(P>0.05)。结论:黄芪甲苷可降低心肌细胞内游离Ca2+浓度,从而减少心律失常的发生,起到抗心律失常的作用。 Objective:To investigate the influence of astragaloside IV on myocardial ischemia arrhythmias in rats about the concen tration of Ca2+ in myocardial cell. Methods: With rat model of ischemic reperfusion injury was prepared, randomly divided into six groups. After 7 days, the concentration of Ca2+ was detected by Fura-2/AM fluorescent probes on the fluorescence spectrometer. Results:There was significant statistic difference with intracellular the concentration of free Ca2+ in model group, compared with the sham group (P〈0.01), and no significant in astragaloside IV with small dose group (P〉0.05), and as well as with the astra- galoside IV medium dose group and large dose group (P〈0.01). And no difference from astragaloside IV with medium and large dose group to propafenone group (P〉0.05). Conclusion: Astragaloside Ⅳ can reduce the concentration of Ca2+ in myocardial in- tracellular, and thus plays a role of anti-arrhythmia.
