六味地黄丸对产后母性应激导致仔鼠生长迟缓的影响

摘要 目的探讨六味地黄丸(TJ-87)对产后母性束缚应激致仔代生长迟缓的影响。方法应用ICR小鼠建立产后母性束缚应激模型,产后第2天将母鼠移离仔鼠,给予母鼠每天1次每次3小时的束缚应激,持续至产后3周断乳时结束。本研究以仔鼠为研究对象:①研究断乳时模型组及正常对照组,观察两组仔鼠的体质量、身长、股骨长、血中胰岛素样生长因子(IGF-1)水平及脑垂体生长激素GH mRNA表达情况;②研究断乳后给予六味地黄丸(TJ-87)大、中、小剂量各治疗组(药物与饲料混合浓度为1.8%,1.2%,0.6%剂量组,连续给药8周)与模型组,观察治疗后各组仔鼠的体质量、身长、股骨长、血中IGF-1水平及脑垂体GH mRNA表达情况。结果①与正常对照组比较,产后3周断乳时模型组仔鼠表现出低体质量、低身长及低股骨长,同时模型组仔鼠血浆IGF-1水平降低,垂体GH mRNA相对表达低下,差异均有统计学意义(P<0.01)。②经六味地黄丸治疗8周后,与模型组比较,六味地黄丸大、中剂量组仔鼠身长及股骨长增加,同时六味地黄丸大、中剂量组仔鼠血浆IGF-1水平升高,其垂体GH mRNA相对表达也升高,差异有统计学意义(P<0.05,P<0.01);六味地黄丸大、中、小剂量组仔鼠体质量差异均无统计学意义(P>0.05)。结论六味地黄丸可能通过GH-IGF-1通路改善产后母性束缚应激仔代的生长。 Objective To explore the effect of " Liuwei Dihuan Pill" on growth retardation induced by postpartum maternal stress in mouse offspring. Methods ICR mice were used to establish the maternal immobilization stress (IS) model, which each dam in the IS group removed from its pups and subjected to IS for 3 hours every day for 3 weeks. (1)The body weight, body length, femur length, IGF-1 level and GH mRNA expression in model group and normal control group were observed. (2)After treatment, the body weight, body length, femur length, IGF-1 level and GH mRNA expression in "Liuwei Dihuang Pill" groups (large, medium and small dose groups administrated with 1.8%, 1.2% and 0.6% medical concentration respectively for 8 weeks) and model group were observed. Results (1) Compared with normal control group, the body weight, body length, femur length were decreased in model group 3 weeks postpartum. The level of IGF-1 and expression of GH mRNA in IS group were lower, with significant difference between the two groups (P 〈 0.01 ). (2) After treated by "Liuwei Dihuang Pill" for 8 weeks, the body length and femur length, IGF- 1 level and expression of GH mRNA in large, medium "Liuwei Dihuang Pill" groups were increased (P 〈0.05, P 〈0.01 ) ; No significant difference was found in body weight among "Liuwei Dihuang Pill" groups (P 〉0.05). Conclusion "Liuwei Dihuang Pili? can improve the growth of postpartum maternal stress in mouse offspring through GH-IGF-1 passageway.
