摘要
目的研究肾虚对大鼠耳蜗蛋白质表达的影响,探讨肾虚发生发展的分子机制。方法幼龄Wistar大鼠94只,雌雄各半,采用缺铁饲料喂养8周,符合肾虚辩证的51只为肾虚组,其余43只为无肾虚对照组。继续喂缺铁饲料8周,取两组大鼠耳蜗膜性组织行labelfree蛋白质组学分析,比较差异蛋白表达。结果与无肾虚对照组比较,肾虚组有47组特异性肽段有显著下调,32组特异性肽段显著上调,肾虚组较对照组的显著性差异肽段有45组,三维图谱分析表明,10 Acb12 actin,beta-like2;784 Actgl Actin,cytoplasmic 2等特异性肽段的表达显著不同,肾虚状态导致两者基峰明显增高。结论肾虚可能通过多种途径引起耳蜗蛋白质特异肽段改变,从而导致肌动蛋白,肌球蛋白减少,细胞骨架结构异常,最终造成听毛细胞静纤毛损伤。
Objective To observe the changes of cochlea protein expression mediated with iron deficiency induced kidney-deficiency and explore the molecular mechanism of kMney deficiency. Methods Ninety four healthy yourlg Wistar ratts were fed witll the iron-insufficient diet for 8 weeks. The modeled rats with kidney deficiency were served as the experimental group (51 rats), and other 43 rats without kidney-deficiency were as the controls. Both groups were continually fed with iron-insufficient feed for another 8 weeks. After treatment, the rats were put into death and cochlea membrane tissues were takeIl to analyze diftereht protein expression by Label-free quantitative proteomic analysis. Results Compared with the control group, 47 groups of specific peptide were down-regulation and 32 group.s of specific peptide were up-regulation in the experimental group. 45 groups of specific peptide were found in the kidney deficiency group significantly different from the control group. Three-dimensional atlas showed that specific peptide (like 10 Acbl2 actin, beta-like 2; 78a Actgl Actin, cytoplasmic 2,et al)expressed differently in 2 groups, the pattern of kidney deficicncy induce base peak raise highly. Conclusions,ions Kidney deficiency may mediate specific Deptide expression through multi-pathway, and decrease actin and myosin expression, and result in an abnormal changes of cytoskeleton structure and finally acoustic hair cell harm.
