不同散粉期胡桃楸离体花粉生活力的测定 被引量:8

摘要 利用TTC法和I-KI法以及离体萌发法对不同散粉期胡桃楸花粉生活力进行测定。结果表明:胡桃楸花粉不适合用TTC法和I-KI法进行测定,而离体萌发法为最佳检测方法。以胡桃楸花粉为外植体,采用蔗糖悬浮溶液为培养基,确定适合胡桃楸花粉萌发的蔗糖浓度为20%。在散粉周期中,散粉盛期花粉具最高生活力。 The pollen viability of Juglans mandshurica during different pollen dispersal periods was tested by means of TI'C, I- KI, and germination in vitro. The results showed that TYC and I-KI methods were not suitable for the pollen viability measurement of Juglans mandshurica, and only in vitro medium, the pollen germination rate could be improved and represent the pollen viability. The pollen was used as explant and the sucrose solution as medium. The germination of the pollen medium of Juglans mandshurica pol- len from Chinese catalpa was observed, and the best sucrose concentration was 20% o It was showed that during the pollen dispersal period, the pollen activity was strongest at the exuberant stage.
