三黄鸡ST6基因的半定量RT-PCR分析

摘要 本研究旨在对三黄鸡ST6(ST6GalⅠ和ST6GalⅡ)基因进行组织表达谱分析。参考鸡ST6GalⅠ和ST6GalⅡ基因序列设计引物,利用半定量RT-PCR进行了组织表达谱分析。结果表明,ST6GalⅠ和ST6GalⅡ基因在各组织均有不同程度的表达,其中ST6GalⅠ在心脏中表达最高,脑中最低;ST6GalⅡ在胸腺中表达最高,在肝脏中表达最低。ST6GalⅠ在各组织中的表达比其相应组织中ST6GalⅡ的表达要低。 The aim of this research was to analyze the expression profile of ST6Gal Ⅰ and Ⅱ (ST6) in Chinese yellow chicken. Based on the sequence of the ST6 of the chicken that reported in NCBI, primers were designed and the Chinese yellow chicken ST6 gene were amplified by PCR. The tissue specific expression profile was analyzed by semi quantitative RT-PCR. The results showed that the ST6 genes were expressed in various tissues but the express levels Varied. The ST6Gal Ⅰ gene has the highest express level in the heart and lowest in brain while the ST6Gal Ⅱ gene was highest in the thymus and lowest in the liver. ST6Gal Ⅰ gene has higher express level than ST6GalⅡ in the same tissue of the yellowchicken.
