摘要
目的:研究六亚甲基二乙酰胺(hexamethylene bisacetamide,HMBA)对人卵巢癌HO8910细胞增殖和细胞周期的影响。方法:采用MTT法测量不同浓度的HMBA(1.0、3.0、5.0、7.0、9.0mmol/L)对人卵巢癌细胞HO8910增殖的影响,流式细胞术观察HMBA作用后细胞周期的变化。结果:MTT显示HMBA处理后的HO8910细胞生长抑制作用随着浓度的增加逐渐增强(r=0.959 8,P<0.05);流式细胞仪检测结果显示,空白对照组HO8910细胞G_1期比例为(67.15±0.57)%,S期为(23.55±0.11)%,G_2期为(9.31±0.48)%;5.0 mmol/L HMBA作用后的HO8910细胞,G_1期比例为(73.64±1.83)%,S期为(17.70±0.74)%,G_2期为(8.66±1.10)%,细胞明显被阻滞于G_1期(P<0.05)。结论:HMBA对人卵巢癌HO8910细胞增殖具有抑制作用。HMBA能将HO8910细胞周期阻滞于G_0/G_1期,诱导HO8910细胞向正常细胞分化,促进细胞凋亡。
Objective:To study the effects of hexamethylene bisacetamide (HMBA) on cell proliferation and cycle in human ovarian cancer HO8910 cells. Methods: The MTT technique was employed to assess the effects of different concentrations of HMBA ( 1.0, 3.0, 5.0, 7.0 and 9.0 mmol/L) on ovarian cancer HO8910 cell proliferation. Changes in cell cycle were determined by flow cytometry. Results: The growth of HO8910 cells was increasingly suppressed as the concentration of HMBA increased ( r = 0. 959 8, P 〈 0.05 ). Flow eytometry showed significant changes in the proportion of cell cycles following administration of 5.0 mmol/L HMBA([67.15±0.57]% vs. [73.64±1.83]% for phaseG1; [23.55±0.11]% vs. [17.70±0.741% for phase S and [9.31± 0.481% vs. [ 8.66 ± 1. 10] for phase G2 ), suggesting that the cell proliferation was inhibited at phase G1. Conclusion: HMBA inhibits proliferation of H08910 cells as a consequence of stagnation at stages G0/G1 and induction of differentiation to normal cells leading to cell apoptosis.
