摘要
Gle1蛋白是一种mRNA核输出因子.最近有研究报道Gle1蛋白参与了酿酒酵母的蛋白质翻译终止过程.为了探讨Gle1蛋白是否在其它生物中也具有同样的功能,本研究利用酵母双杂交、免疫共沉淀以及免疫共定位等方法证实了人Gle1蛋白与参与蛋白质合成终止的两类肽链释放因子均能够相互作用,提示hGle1蛋白可能在人细胞中参与了蛋白质的翻译终止过程.进一步在HeLa细胞中利用双荧光素酶报告系统分析人Gle1蛋白对蛋白质翻译终止的影响,结果显示,hGle1的过表达能促进细胞内蛋白质的翻译终止.为进一步探讨hGle1蛋白在蛋白质合成中的作用机制奠定了基础.
Glel is required for mRNA export in yeast and human cells. It was reported that Glel is als, involved in efficient translation termination in Saccharomyces cerevisiae. However, whether Glel plays role in translation termination in other organisms has not been determined. In this study, the interactio between Glel and polypeptide release factors from human cell was identified by yeast two-hybrid screen co-immunoprecipitation and co-localization analysis. The results showed that hGlel interacted witl translation termination factors heRFl and heRF3, suggesting its participation in the human translation termination. The regulatory function of hGlel in translation termination was investigated in HeLa cell using dual-luciferase reporter system. The results indicated that the overexpression of hGlel increased th translation termination activity in a dose dependent manner. The results provide important implication fo further studies of the molecular mechanisms of hGlel in translation.
