摘要
为获得兔眼蓝莓高效快繁的技术体系,以杰兔幼嫩茎段为试验材料,对增殖培养基、叶片再生体系、生根培养基、炼苗基质等组培苗生产的几个关键因素进行了优化.结果表明,WPM(改良)+1.0 mg/L ZT增殖培养基能使新梢增殖率达6倍以上,丛生苗生长速度快,而且株型健壮.在无菌苗叶片离叶尖1/3处,沿垂直叶片主脉方向将叶片切成二部分,叶片背面朝下接种于WPM(改良)+1.7 mg/L ZT培养基中,先暗培养20 d后光照培养,结果叶尖端和叶柄端的再生率分别达到93.8%和79.5%;1/4 WPM(改良)+1 g/L活性碳+0.1 mg/L IBA生根培养基能使试管苗生根率达95%;以水苔藓为炼苗基质可使幼苗成活率高达100%.因此,本研究从叶片再生、增殖培养、生根培养到炼苗这一系列环节进行了优化,建立了一种可高效、快速、稳定获得优良的兔眼组培种苗的技术.
To investigate a fast and efficient propagation technology of Vaccinium ashei Reade, the key technica1 factors, such as proliferation culture medium, leaf regeneration system, rooting culture mediums, and seedling adaptation medium, were optimized when young stem segments of Premier were used as test material. The results indicated that shoot proliferation rate increases more six times, multiple shoots are growing fast, and plant type is robust, when WPM (improved) + 1.0 mg/L ZT proliferation medium was used. Sterile leaves of seedlings were cut into two portions in 1/3 from the tip of the leaves and along the vertical direction of the main veins of leaves, and the cut leaves with back down manner inoculated in the WPM(improved) + 1.7 mg/L ZT medium with the side down, first cultured in the dark for 20 d, and then cultured in the 15 h lighting and 9 h darkness, the results showed that the regeneration rate of leaf tip and petiole end were 93.8% and 79.5%, respectively. Rooting grate can reach 95% in 1/4 WPM(improved) + 1 g/L carbon + 0.1 mg/L IBA rooting medium, seedlings survival rate can be up to 100%, when sphagnum moss was used as seedling adaptation medium. Thus, the key technologies of tissue culture seedlings production to Vaccinium ashei Reade were obtained in this study, and an excellent technology system of Vaccinium ashei Reade tissue culture seedlings sue culture seedlings of Vaccinium ashei Reade efficiently, was successfully established, rapidly, and stably. which could produce tissue culture seedlings of Vaccinium ashei Reade efficiently, rapidly, and stably.
