摘要
据国外发表的人致病性大肠杆菌 1型及P型菌毛蛋白结构基因 (pilA及papA)序列 ,设计并合成了分别位于PilA及papA保守区的 2对引物。经PCR从 1株带有甘露糖敏感血凝特性 (MSHA)及甘露糖抵抗血凝特性 (MRHA)的鸡致病性大肠杆菌 (HJ2 0e)染色体中扩增到大小分别在 6 5 7bp及 5 41bp的 2种阳性产物。通过核酸序列测定分别确证所扩增的DNA片段分别为pilA及papA基因。经酶切、连接、转化和筛选 。
According to the published type 1 and P pili structure gene (pilA and papA)sequence from human Escherichia coli,two pairs of primers were designed and synthesized,which localize conservative regions of pilA or papA gene,657bp and 541bp DNA fragments were generated by polymerse chain reaction(PCR)from an avian Escherichia coli (HJ 20 ,O 114 )strain chromosomal.The nucleotide sequences of two DNA fragments were analysed,the results showed that these DNA fragments belong to type 1 and P pili protein structural gene(pilA and papA).pilA and papA were cloned into pGEM4 and puC19 plasmids respectively by enzyme digest,ligation,transformation and screen.
