摘要
为研制一种快速、简便的胶体金免疫层析法的试纸条,用于对动物源食品中氟喹诺酮类药物残留检测,研究以沙拉沙星(SALX)多克隆抗体与胶体金结合为金标抗体,将纯化好的金标抗体喷涂在玻璃纤维垫上;包被抗原SALX-OVA和山羊抗兔二抗分别包被在硝酸纤维素膜上,制成免疫层析快速检测试纸条,并对试纸条的性能进行测定。结果表明,该快速检测试纸条的灵敏度为30ng/ml,检测时间为5~10min,不但可对沙拉沙星残留进行检测,而且还可对诺氟沙星、环丙沙星和恩诺沙星的残留进行检测。表明试纸条可对氟喹诺酮残留进行多元快速检测。
To develop a rapid and simple method to detect fluoroquinolones residues in animal-derived food, colloidal gold immunochromatography test strips were prepared. The colloidal gold were labeled with the anti-SALX polyclonal antibody, and then it was sprayed onto the glass fiber. The SALX-OVA coating antigen and goat anti-rabbit IgG were bound onto a nitrocellulose membrane respectively, thus to prepare immunoehromatographic strips and test its performances. The detection limit of the test strips was 30g/ml. The test could be completed within 5 × 10min and the strips could detect not only sarafloxaein residues, but also norfloxacin, ciprofloxacin and enrofloxacin residues. The colloidal gold test strip can be employed for rapid and multivariate detection for the residues of fluoroquinolones.
