骨髓间充质干细胞复合载异补骨脂素支架材料修复骨缺损的研究 被引量:6

摘要 目的:探讨异补骨脂素对骨髓间充质干细胞成骨分化的促进作用及骨髓间充质干细胞复合载异补骨脂素支架材料应用于修复兔下颌骨缺损的可行性。方法:36只新西兰大白兔随机分为3组并制备双侧下颌骨缺损模型,A组为实验组,缺损区内植入BMSCs复合载异补骨脂素支架材料;B组为对照组,缺损区内植入BMSCs复合羟基磷灰石材料,C组为空白组,缺损区内不植入任何材料。分别于术后2、4、8、12周处死各组动物,每次9只,取双侧下颌骨制备组织标本,行大体观察、影像学分析、HE染色、扫描电镜检测,并对影像学分析值及成骨情况加以评价,对分析数据结果使用SPSS17.0统计软件进行分析有统计意义(P<0.05)。结果:影像学检查及组织学染色结果显示A组骨缺损处愈合程度,成骨速度及其质量明显优于明显优于其他两组;扫描电镜显示A组复合材料与组织相容性好,组织间无炎症刺激反应;并通过统计各组各期牙CT分析数据及新生骨面积,A组的成骨情况也明显优于B、C组(P<0.05)。结论:BMSCs复合载异补骨脂素支架材料具有明显骨诱导作用,是一种新型复合材料,可望成为临床应用中修复颌骨缺损的新型材料。 Objective:To investigate the psoralen on bone marrow mesenchymal stem cell osteogenic differentiation and promote the role of bone marrow mesenchymal stem cells combined with scaffolds containing psoralen used in repairing mandibular defects in rabbits feasibility.Methods:36 New Zealand white rabbits were randomly divided into 3 groups and prepared bilateral mandibular defect model.A group was experimental group with defect area implanted BMSCs composite containing psoralen scaffold.B was the control group with the defect area implanted BMSCs hydroxyapatite composite materials.C group was the control group,the area was not implanted with any material.After 2,4,8,12 weeks,the animals were sacrificed with 3 in each group.Bilateral mandibule was preparaed to take tissue samples,gross observation,image analysis,HE staining,scanning electron microscopy observation.The value of image analysis and osteogenic conditions were evaluated.Results:Image and histological staining showed that the bone defect healing in group A was better than the other two groups.Scanning electron microscopy showed that composite material of group A was compatible,the organization inflammatory response without a stimulus;and through the group stages of statistical analysis of dental CT data and new bone area,A group of bone situation was significantly better than the other two groups.Conclusion:BMSCs composite scaffolds containing psoralen obvious bone induction is a new composite material,which can be expected to become the clinical application for mandibular defects.
