臭氧氧化预处理对大鼠肾I/R组织Toll样受体4表达的影响 被引量:2

Effect of Ozone Oxidative Preconditioning on Expression of Toll-like Receptor 4 in Renal Ischemia Reperfusion Injury in Rats

摘要 目的观察臭氧氧化预处理(OzoneOP)对大鼠肾缺血再灌注损伤(ischemia-reperfusion injury,I/R)组织内Toll样受体4(TLR4)表达的影响。方法将48只大鼠随机分为假手术组、假手术+OzoneOP组、I/R组、I/R+OzoneOP组,每组各12只。术前15d始对包含OzoneOP的大鼠经直肠吹入氧气和臭氧的混合气体5~5.5 ml(臭氧浓度50 mg/L,1 mg/kg,每日1次),之后按分组要求建立原位大鼠单侧肾I/R动物模型。24 h后检测尿素(BUN)、肌酐(Cr)、TLR4 mRNA表达、TLR4蛋白表达情况。结果 I/R组和I/R+OzoneOP组BUN、Cr、TLR4 mRNA含量、TLR4蛋白含量明显高于假手术组和假手术+OzoneOP组,差异有统计学意义(P<0.05),而I/R组上述4项指标又明显高于I/R组+OzoneOP组,差异均有统计学意义(P<0.05),而假手术组与假手术+OzoneOP组比较差异无统计学意义(P>0.05)。结论 OzoneOP可以减轻肾I/R,其机制可能通过抑制TLR4的表达而发挥作用。 Objective To observe effects of ozone oxidative preconditioning(OzoneOP) on expression of toll-like receptor 4(TLR4) in rats with renal ischemia-reperfusion injury(I/R).Methods Forty-eight rats were randomly divided into sham operated group,sham operated+OzoneOP group,I/R group and I/R+OzoneOP group(n=12 in each group).Rectums of rats with OzoneOP were insufflated Oxygen+Ozone mixed gas 5~5.5 ml(Ozone concentration 50 mg/L,1 mg/kg,1 time a day) 15 days before operation,then the rats' unilateral renal I/R animal models were set up according to requirements of each group.The blood urea nitrogen(BUN),creatinine(Cr),expressions of TLR4 mRNA and TLR4 protein were detected 24 hours later.Results BUN,Cr,contents of TLR4 mRNA and TLR4 protein in I/R group and I/R+OzoneOP group were significantly higher than those of sham operated group and sham operated+OzoneOP group(P0.05),while the above 4 indexes in I/R group were significantly higher than those of I/R+OzoneOP group(P0.05);but the differences between sham operated group and sham operated+OzoneOP group were not statistically significant(P0.05).Conclusion OzoneOP can alleviate renal I/R injury in rats by way of the inhibition of TLR4 expression.
