Plackett-Burman设计在筛选影响发芽芸豆蛋白酶活性的金属离子中的初步应用

摘要 为通过发芽处理提高奶花芸豆的商品价值,需要确定影响发芽奶花芸豆蛋白酶活性的金属离子的种类。通过Plackett-Burman试验设计方法,结合紫外光谱法测定蛋白酶活性,就Na+,K+,Mg+,Cu2+,Ca2+,Zn2+对发芽芸豆蛋白酶活性的影响进行考察。试验表明,Cu2+对蛋白酶活性有显著负效应影响(p<0.05),Zn2+对蛋白酶活性有显著正效应影响。 Kidney bean (Phaseolus vulgaris) was sprouted to improve the value of commodity. Determined the influence of the metal ions on proteinase activity of germinating kidney bean. The protease activity was detected by Plackett- Burman experimental design methods combined with UV spectrometry, investigated the influence of Na+, K+, Mg+, Cu2+, Ca2+, Zn2+ on the protease activity of generating kidney bean. Experiments showed Cu2+, Zn2+ have a significant effect (p 〈0.05) on protease activity, Cu2+ had a negative effect on protease activity and Zn2+ had a positive effect.
