黄连解毒汤对代谢综合征大鼠心肌的保护作用及对胰岛素信号转导的影响 被引量:5

Effects of Huanglian Jiedu decoction on myocardium and signal transduction of insulin receptor substrate in rats of metabolic syndrome

摘要 目的观察黄连解毒汤(HLJDT)对代谢综合征(MS)大鼠心肌的保护作用及对胰岛素信号转导的影响,探讨其保护心肌的分子机制。方法高糖、高脂及高盐饲料及20%高蔗糖水喂养(HF饮食)喂养Wistar大鼠建立MS大鼠模型,以HLJDT干预治疗12 w,检测血糖、血清胰岛素和血脂水平,Western印迹方法检测HLJDT治疗后MS大鼠心肌组织内核因子-κB(NF-κB)、磷酸化Akt(P-Akt)蛋白表达及胰岛素受体底物1(IRS-1)蛋白色氨酸磷酸化水平。结果 HLJDT治疗MS大鼠后,心肌组织内NF-κB表达及IRS-1丝氨酸磷酸化水平较MS组下降,P-AKT显著增加。结论黄连解毒汤通过抑制炎症降低IRS-1丝氨酸磷酸化水平,这可能是其保护心肌的机制之一。 Objective To investigate the effects of Huanglian Jiedu decoction (HLJDD) on myocardium and signal transduction of insulin receptor substrate in rats of metabolic syndrome (MS) , and explore its possible molecular mechanism of improving myocardial lesions. Methods MS rats were established with obese-diets (a high-fat, high-glucose and high-sah diet and sucrose solution). MS rats in the testing group were treated with HLJDD for 12 weeks. Fasting serum glucose, insulin and blood-fat were determined, and Western blot was performed to analyze the proteins in signal transduction pathway, including nuclear factor kappa B (NF-κB) , phosphorylation Akt and tyrosine phosphorylation levels of insulin receptor substrate-1 ( IRS-1 ). Results NF-KB protein and tyrosine phosphorylation and IRS-1 were decreased, and phospho-AKT protein was increased significantly in MS rats treated with HLJDD, compared with untreated MS rats. Conclusions HLJDD could protect myocardium from IR-mediated injury by inhibiting IRS serine phosphorylation catalyzed by inflammation in MS rats.
