摘要
目的:研究疏肝饮及其组分对反复避水应激(RWAS)大鼠内脏痛觉过敏(VHL)的调节作用,并探讨疏肝饮是否通过调节背根神经节(DRG)中瞬时受体电位通道香草酸受体1(TRPV1)和结肠组织中P物质(SP)表达起作用。方法:应用RWAS法复制具有VHL特点的肠易激综合征(IBS)大鼠模型,观察疏肝饮及其组分对结直肠扩张(CRD)引起大鼠腹壁收缩反射(AWR)压力阈值的影响;分别应用免疫组化法和Real-time PCR法检测大鼠结肠组织中SP和DRG中TRPV1的表达变化。结果:模型大鼠引起AWR的压力阈值明显低于正常对照组,疏肝饮可以使模型大鼠引起AWR的压力阈值明显升高;疏肝饮及其组分可以降低模型大鼠结肠组织中高表达的SP和DRG中高表达的TRPV1。结论:疏肝饮可以改善RWAS大鼠的VHL,其作用机制与其调节结肠组织中SP和DRG中TRPV1的表达有关。
Objective :To study the regulatory effect of Shuganyin and its constituents on visceral hyperalgesia(VHL) in the repeated water avoidance stress(RWAS) rats and investigate whether they exert their action by modulating the expres- sion of transient receptor potential channel vaniltoid receptor 1 ( TRPV1 ) in dorsal root ganglion(DRG) and the expression of substance P(SP) in colon~ Methods :We observed the effect of Shuganyin and its constituents on the threshold value of abdominal withdraw reflex (AWR) induced by colorectal distension (CRD) in irritable bowel syndrome (IBS) rat model which was produced by RWAS and charactered with VHL. The expression of SP in colon was measured by immunohisto- chemistry while the expression of TRPV1 mRNA in DRG was measured by Real -time PCR. Results :The threshold value of AWR in model rat was lower than that of control rat. Shuganyin can enhance the threshold value of AWR in model rat. Shuganyin and its constituents can reduce the level of SP expressed in colon and the level of TRPV1 expressed in DRG. Conclusion : Shuganyin can improve VHL in RWAS rats by regulating the expression of SP in colon and the expression of TRPV1 in DRG.
