摘要
目的:建立二氢吡啶类钙拮抗剂非洛地平和尼莫地平对映体的手性分离方法。方法:在Pirkle型Whelk-O1手性柱上分别考察在流动相正己烷中,不同的醇类添加剂及碱性添加剂对手性分离的影响。结果:当流动相为正己烷-乙醇-二乙胺(85∶15∶0.1,v/v/v),流速0.5 mL·min-1,温度25℃时非洛地平和尼莫地平分离度R分别为1.77和1.45。结论:本法操作简便,可用于非洛地平和尼莫地平对映体的分离分析。
Objective:To establish an HPLC chiral separation method for dihydropyridine calcium antagonists felodipine and nimodipine enantiomers.Methods: The chiral separation of felodipine and nimodipine was studied on(R,R)-Whelk-O1 chiral columns,taking hexane as the mobile phase with alcohol and alkaline additives of different types and concentrations.Results:On(R,R)-Whelk-O1 chiral column,felodipine and nimodipine could be baseline separated using hexane-ethanol-diethylamine(85∶ 55∶ 0.1,v/v/v) as the eluent at a flow rate of 0.5 mL·min-1 and a temperature of 25 ℃.Conclusion:It is recommended that this simple and selective method could be used for enantioseparation of felodipine and nimodipine enantiomers.
