摘要
目的探讨高脂血症大鼠肾脏内热休克蛋白70(HSP70)的表达变化。方法 10只健康大鼠适应性喂养一周后随机分成正常组和高脂血症组。12周后取血,检测血脂;取肾组织分别进行HE染色和HSP70免疫组织化学染色,应用计算机图像分析系统检测HSP70的平均光密度变化。结果血脂检测结果显示:正常组TG、TC和HDL-C与高脂血症组存在差异。HE染色结果显示:高脂血症组部分肾小管上皮细胞破碎,核固缩,胞浆轻微肿胀变性,细胞排列紊乱。肾小囊周围有少量炎性细胞浸润,肾小囊内细胞有空泡形成。免疫组化结果显示:高脂血症组肾小管HSO70表达强于正常组。图像分析结果显示:与正常组HSP70的平均光密度值(0.1185±0.0078)比较,高脂血症组HSP70平均光密度值(0.3106±0.0484)增高,有统计学差异(P<0.05)。结论高脂血症可导致大鼠肾组织内HSP70表达增高,高脂血症大鼠肾组织形态学损伤与HSP70含量的增高具有一致性。
Objective To investigate changes of the expression of heat shock protein 70(HSP70)in hyperlipidemic rats induced by high fat diet and and its effect on lipid renal damage.Methods Ten rats were fed with common chow for one week,then divided into normal group and hyperlipidemia group randomly,and the level of blood-fat of the rats was measured after twelve weeks.The renal tissues were observed by using HE staining and HSP70 was detected by immunohistochemistry.The value of average optical density of HSP70 was measured by the image analyzing system of the computer.Results There are differences between normal group and hyperlipidemia group in the level of TG,TC and HDL-C.HE staining results showed that a few of renal tubular epithelial cells of hyperlipidemia rats broken,disordered,appeared karyopyknosis.and their cytoplasm became slight swelling and degeneration.A few inflammatory cells infiltrated around the renal capsules while the renal tubules showed varying degrees of vacuolar degeneration.Immunohistochemistry staining results showed that expression level of HSP70 was higher in hyperlipidemia group than in normal group.The average optical densities of HSP70 had significant difference between normal group and hyperlipidemia group(P0.05).Conclusion Hyperlipidemia can upregulate the level of HSP70 in renal organs,the renal injury in hyperlipidemic rats is consistent with elevating of the HSP70 level.
