摘要
建立了中成药中多种蒽醌类化合物(芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚)的含量测定方法,为中国药典中含蒽醌的中成药提供一致的测定方法。色谱柱Agela Venusil XBP-C18柱(4.6mm×250mm,5μm);流动相为乙腈-0.1%磷酸溶液,梯度洗脱;检测波长为226nm;流速为1.0mL.min-1;柱温为40℃。芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚分别在0.040—1.200、0.010—1.200、0.040—1.200、0.040—1.200、0.020—0.600μg范围内呈良好的线性关系,相关系数分别为0.9998、0.9997、0.9991、0.9997、0.9993,平均回收率(n=9)分别为99.26%—102.18%(RSD1.68%—2.64%)、98.85%—99.61%(RSD 1.02%—1.94%)、100.29%—104.65%(RSD 1.90%—2.57%)、97.68%—98.68%(RSD 1.47%—2.53%)、96.18%—100.21%(RSD 1.57%—2.82%)。方法快速简便,重复性好,测定结果准确可靠,能够用于大黄剂中蒽醌类衍生物的含量测定。
The method for determination of the contents of various anthraquinone derivatives(aloeemodin,rhein,emodin,chrysophanol and physcion) in Chinese patent drug was established,that provided the consistent determination method for Chinese patent drug containing anthraquinone in Chinese Pharmacopoeia.The compounds were separated by Agela Venusil-C18 column(4.6mm×250mm,5μm) with mobile phase of acetonitrile-0.1% phosphoric acid for gradient elution at a flow rate of 1.0mL.min-1,the detection wavelength was 226nm,and column temperature was 40℃.Aloeemodin,rhein,emodin,chrysophanol and physcion had good linear relationships in the ranges of 0.040—1.200,0.010—1.200,0.040—1.200,0.04—1.200,0.020—0.600μg,and the correlation coefficients of them were respectively 0.9998,0.9997,0.9991,0.9997 and 0.9993,the average recoveries(n=9)of which were 99.26%—102.18%,98.85%—99.61%,100.29%—104.65%,97.68%—98.68%,96.18%—100.21%,respectively,and the RSDs were respectively 1.68%—2.64%,1.02%—1.94%,1.90%—2.57%,1.47%—2.53% and 1.57%—2.82%.The method is fast,simple with good repeatability and accurate results,that can be applied to determination of contents of anthraquinone derivatives in rhubarb preparations.
