摘要
目的:观察三丫苦对高脂饮食性胰岛素抵抗大鼠GLP-1mRNA的影响。方法:40只SD雄性大鼠随机分为正常对照组、模型对照组、三丫苦组、罗格列酮组,建立高脂饮食性胰岛素抵抗大鼠模型。检测各组大鼠TC、TG、FBG、FINS等指标的变化,清醒状态下高胰岛素-正葡萄糖钳夹实验检测葡萄糖输注率,Real-time PCR法检测肝脏组织GLP-1mRNA的表达。结果:与正常对照组比较,模型对照组TC、TG、FBG、FINS升高(P<0.01),葡萄糖输注率降低(P<0.01)。给予三丫苦后,大鼠TC、TG、FBG、FINS降低(P<0.01或P<0.05),GIR水平升高(P<0.05),与罗格列酮组比较TC和TG降低(P<0.05)。与正常对照组比较,模型对照组GLP-1mRNA表达显著降低(P<0.01)。与模型对照组比较,三丫苦组和罗格列酮组GLP-1mRNA表达均有不同程度的升高(P<0.01或P<0.05)。与罗格列酮比较,三丫苦组GLP-1mRNA表达升高(P<0.05)。结论:三丫苦对高脂饮食性胰岛素抵抗大鼠糖脂代谢及GLP-1mRNA有一定调节作用。
Objective : To observe the effect of Sanyaku on GLP - 1 mRNA of insulin - resistance rats induced by high - fat diet. Method :40 male SD rats were divided into 4 groups ( normal control group, model control group, Sanyaku group and Rosiglitazone group) randomly. The insu- lin - resistance rat models induced by high - fat diet were established. The change of the index TC, TG, FBG, FINS of each group was examined. The glucose infusion rate (GIR) was examined by hyperinsulinemic - euglycemic clamp when the rats were in consciousness. The expression of GLP - lmRNA in liver tissue were detected by Real -time PCR method. Result:The TC ,TG,FBG and FINS were obviously higher and the GIR were obviously lower in the model control group than in the normal control group (P 〈 0.01 ). After given Sanyaku, the rats'TC, TG, FBG and FINS decreased ( P 〈 0. 01 or P 〈 0.05 ) and the GIR increased ( P 〈 0.05 ). Compared with TC and TG of the Rosiglitazone group, that of the Sanyaku group decreased (P 〈 0.05 ). Compared with the expression of GLP - 1 mRNA of the normal control group, that of the model control group de- creased obviously (P 〈 0. 01 ). Compared with the expression of GLP- 1 mRNA of the model control group, that of the Sanyaku group and the Ros- iglitazone group increased at different degrees (P 〈 0. 01 or P 〈 0.05 ). Compared with the expression of GLP - lmRNA of the Rosiglitazone group, that of the Sanyaku group increased ( P 〈 0.05 ). Conclusion : Sanyaku has regulating effect on glucose and lipid metabolism and GLP - 1 mRNA of insulin- resistance rats induced by high- fat diet.
