去甲肾上腺素诱导ConA活化的大鼠淋巴细胞产生应激免疫抑制蛋白 被引量:6

摘要 目的 :观察去甲肾上腺素在体外诱导正常大鼠淋巴细胞产生应激免疫抑制蛋白。方法 :采用体外培养的方法 ,以ConA活化正常大鼠的淋巴结细胞 ,同时加入去甲肾上腺素共同孵育 12~ 16h ,测定细胞提取物对正常小鼠淋巴细胞转化的影响。并利用应激免疫抑制蛋白单克隆抗体以抗体中和及ELISA法 ,鉴定应激免疫抑制蛋白。结果 :去甲肾上腺素 (10 -5~ 10 -3 mol·L-1)可在体外诱导以ConA活化的淋巴细胞产生一种能抑制正常淋巴细胞转化的物质。用应激免疫抑制蛋白单克隆抗体所作的抗体中和及ELISA测定发现 ,提取物中的抑制物可被应激免疫抑制蛋白单克隆抗体识别。结论 Objective: To establish a cellular model for generation of immunosuppressive protein induced by norepinephrine from normal lymphocytes in vitro . Methods: Normal lymphocytes activated by Con A were incubated with norepinephrine. Extract of lymphocyte was made after the incubation and the effect on normal lymphocyte proliferation was tested. Monoclonal antibody against immunosuppressive protein was used to identify the extract. Results: Lymphocyte proliferation in response to ConA was significantly suppressed by the extract. The suppression induced by the extract was partially reversed by the monoclonal antibody against immunosuppressive protein . ELISA results showed that there were molecules in the extract, which were combined by the monoclonal antibody. Conclusion: Immunosuppressive protein could be induced by norepinephrine from ConA activated lymphocytes.
