摘要
目的研究前列腺素E1脂微球载体制剂(Lipo-PGE1)联合肾衰丸对慢性肾衰竭大鼠肾脏的保护作用,并初步探讨其作用机制。方法 5/6肾切除法复制大鼠慢性肾衰模型。将大鼠随机分为假手术组(10只)、模型组(10只)、Lipo-PGE1治疗组(10只)、肾衰丸治疗组(10只)以及Lipo-PGE1联合肾衰丸治疗组(10只),8周后检测各组大鼠血尿生化指标,观察肾组织病理改变;免疫组织化学方法检测各组大鼠肾小球纤维连接蛋白(FN)、转化生长因子-β(1TGF-β1)和结缔组织生长因子(CTGF)的蛋白表达水平。结果模型组大鼠尿蛋白、血清尿素氮和肌酐水平显著高于假手术组,肾小球系膜区和肾间质纤维化病变显著。Lipo-PGE1治疗组、肾衰丸治疗组以及Lipo-PGE1联合肾衰丸治疗组尿蛋白、血清尿素氮和肌酐水平较模型组均有不同程度的下降,肾脏的病变有明显的减轻,而Lipo-PGE1联合肾衰丸治疗组上述指标下降更为明显。与假手术组相比,模型组大鼠肾皮质FN、TGF-β1和CTGF的表达明显上调。Lipo-PGE1治疗组、肾衰丸治疗组以及Lipo-PGE1联合肾衰丸治疗组,FN、TGF-β1和CTGF的表达均有不同程度的抑制,而Lipo-PGE1联合肾衰丸治疗组抑制程度更为显著。结论 Lipo-PGE1联合肾衰丸能更有效地减少慢性肾衰竭大鼠蛋白尿,保护肾功能,延缓肾脏纤维化的进展,具有明显的肾脏保护作用。
【Objective】 To investigate the effect of lipo-prostaglandin E1（prostaglandin E1 incorporated in lipid microspheres,Lipo-PGE1） with Shenshuai pill on the renal injury in chronic renal failure rats and its possible mechanism.【Methods】 50 male rats were divided into 5 groups at random： CRF（chronic renal failure） group（n =10）,Lipo-PGE1 treated group（n =10）,Shenshuai pill treated group（n =10）,Lipo-PGE1 with Shenshuai pill treated group（n =10）,and sham operation group（n =10）.The animal model of chronic renal failure was induced by 5/6 nephrotomized.8 weeks after the treatment,blood ureanitrogen（BUN）,creatinine（Cr） and 24 hour urinary protein were detected in the end of experiment.The pathology of the rats was also observed through lightmicroscopy.The expressions of extracellularmatrix fibronectin（FN）,transforming growth factor-β1（TGF-β1）,and connective tissue growth factor（CTGF） were analyzed by immunohistochemistry.【Results】 The 24 hour urinary protein,serum creatinine,serum urea nitrogen in Lipo-PGE1 treated group,Shenshuai pill treated group,and Lipo-PGE1 with Shenshuai pill treated group were significantly reduced,while the co-treated group was lower than that in Lipo-PGE1 and Shenshuai pill treated group.And the expressions of FN,TGF-β1,and CTGF were also obviously inhibited in Lipo-PGE1 group,Shenshuai pill treated group and co-treated group,which that co-treated group was more significant than that in Lipo-PGE1 and Shenshuai pill treated group.【Conclusions】 Lipo-PGE1 with Shenshuai pill may play an important role in the CRF,which may improves chronic renal failure rats renal function,inhibits the proliferation of glomerular cells and reduces the deposition of extracellular matrix.
