摘要
                
                    采用细胞破碎、硫酸铵分级沉淀和Sephadex G-75凝胶过滤的方法,对黑曲霉(Aspergillus niger)J6产生的有机磷农药降解酶进行分离和纯化,并对其酶学性质进行研究。结果显示:黑曲霉J6有机磷农药降解酶定位在细胞膜上,其分子量为66 000。该降解酶最适pH值为7.0,最适反应温度为70℃,Fe3+、Cu2+、Li+、Zn2+、Mg2+、NaN3、EDTA、SDS、PSMF对该降解酶有不同程度的激活作用,而Mn2+对其有轻微的抑制作用,Ca2+和Triton对其有强烈的抑制作用。
                
                A organophosphorus hydrolase from strain Aspergillus niger J6 was isolated and purified by means of cell disruption, ammonium sulfate precipitation and Sephadex G-75 chromatography, and its enzymatic properties were studied. The results showed that the degradation enzyme was located on the cell membrane, with its molecular weight being 66 000 by SDS-PAGE. The optimal reactive temperature and pH of the degradation enzyme were 70 ℃ and pH 7.0. Some metal i- ons( Fe3+, Cu2+, Li+, Zn2+, Mg2+) and chemical reagents( NAN3, EDTA, SDS and PSMF) showed activation on the deg- radation enzyme, while Mn+ displayed slight inhibition, and Ca2+ Triton had strong inhibition.
