摘要
目的:用雌激素受体(Estrogen Receptor,ER)表达阳性细胞株人乳腺癌细胞MCF-7检测淫羊藿苷及其拟代谢物的雌激素样活性[1]及构效关系。方法:用MTT法,增殖细胞核抗原(Proliferating CellNuclear Antigen,PCNA)免疫组化法检测其对体外培养MCF-7细胞增殖及细胞周期的影响;Real-timeRT-PCR法检测ERα、ERβ和PS2 mRNA表达水平变化。结果:淫羊藿素促进MCF-7增殖,而淫羊藿苷、淫羊藿次苷I和脱水淫羊藿素促增殖效应不明显;淫羊藿素10-9mol.L-1组PCNA阳性表达强于其它三组;淫羊藿素极显著增加ERα和PS2 mRNA的表达水平。所有受试物均不促进ERβmRNA表达。淫羊藿素促进ERα蛋白表达强于其它三组。结论:淫羊藿素有明显的雌激素样作用,而淫羊藿苷及其它两种受试物雌激素样作用不明显。四种受试物均不促进ERβmRNA表达。
Objective ： To investigate the effect of estrogen - like activities of icariin and its possible metabolites by ER - positive cell line MCF -7 for human breast cancer cell. Methods： MTT method and PCNA immunohistochemistry were performed to investigate the effects of ieariin, icaritin, icariside I and anhydroicaritin on MCF -7 proliferation and cell cycle progression in vitro. Real - time RT - PCR method was used to detect ERa, ERβ and PS2 mRNA expression lev- els. Resuhs：Icaritin promotesd the proliferation of MCF- 7; icaritin 10-9 mol · L^-1 had a stronger expression of PCNA than other three;icariin, icariside I and anhydroicaritin showed no significant effect on proliferation; icaritin significantly increased the expression of ERa and PS2 mRNA levels. However, all the subjects didnt promote the expression of ERβ mRNA. Conclusion ： Icaritin has a stong estrogen - like effect, but other three subjects are not obvious. Icaritin with the strongest estrogen -like activity of four subjects do not promote the ERβ mRNA expression.
