醇类沉淀法提取尿液蛋白的优化策略 被引量:4

摘要 目的研究提取条件,探讨提取尿液蛋白的优化策略。方法收集健康志愿者的混合尿液标本,采用甲醇、乙醇、异丙醇沉淀法在不同的条件下提取尿蛋白,这些条件包括:醇类试剂的种类、醇类试剂的比例、醇类试剂与尿液作用的不同温度及作用时间、不同的温度处理程序等。然后测定提取的蛋白浓度,评价蛋白回收量;十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)评价总蛋白条带;蛋白免疫印迹评价在不同条件下提取的目的蛋白条带。结果孵育条件固定为4℃30 min时,溶剂为75%乙醇可提取蛋白最多为(649.03±28.33)μg;SDS-PAGE和免疫印迹结果显示,在不同醇类溶剂条件下提取的总蛋白条带和目的蛋白条带呈趋势变化。提取试剂条件固定为75%乙醇时,改变孵育的温度方案,显示先4℃孵育30 min后,再-20℃过夜提取的总蛋白最多为(599.93±42.20)μg;SDS-PAGE和免疫印迹结果都显示,孵育温度方案不同,所提取的总蛋白条带和目的蛋白条带无明显差异。结论 75%乙醇,在4℃孵育30 min后,-20℃过夜条件下,可高效提取尿中胰淀粉酶(AMY),用于尿AMY定性定量检测。醇类溶剂的比例对提取的总蛋白量影响很大,且对不同分子量蛋白有选择性;孵育温度条件主要影响总蛋白量。
