胶囊渗透压泵植入小鼠皮肤创面持续给药模型的建立 被引量:1

Establishment of a novel mice excisional wound model using sustained drug delivery system

摘要 目的制作小鼠全层皮肤切除创面后通过植入胶囊渗透压泵而建立皮肤创面持续性给药小鼠模型。方法在36只C57BL/6小鼠肩胛间区制作全层皮肤切除创面后,将小鼠随机分为3组,每组12只,根据有无植入渗透压泵及泵植入部位不同将各组分别命名为对照组(无泵植入)、皮下组及腹腔内组并行相应处理。采用大体观察及组织学检验的方法对术后各组创面愈合情况进行分析,观察比较不同部位植入的泵对小鼠一般状况及皮肤创面修复的影响。结果植入泵在术后对小鼠全身状态无明显影响。皮下组和腹腔内组小鼠创面愈合情况与对照组相较差异无统计学意义(P>0.05)。结论胶囊渗透压泵植入给药的小鼠全层皮肤切除模型较旧有各给药创面愈合模型更具持续性及量化、可控性,可为针对创伤愈合机制及药物作用所进行的研究提供依据。 Objective Establish a novel full thickness exeisional wound mice model with sustained drug delivery system by implanting a osmotic pump. Methods With full thickness excisional wound created in mid-scapular area, 36 C57BL/6J mice were randomly divided into three groups. Mice without pump served as control group. Mice received pump implantation were grouped depending on the sites of the implants (intraperitoneal and subcutaneous group ). Wounds were observed and evaluated. Histology, immunohistochemistry and immunofluorescence were performed in order to investigate if the implanted pump will affect general condition, survival quality and wound healing process of the mice. Results Implanted pump did not cause significant discomfort in post-operative mice. No significant difference was found in the wound healing process among intraperitoneal and subcutaneous groups ( P 〉 0.05 ). Conclusions Compared with existing model, the excisional wound model with implanted osmotic pump is more persistent, accurate, quantifiable and controllable. It can be considered as a model for researches concerning wound healing mechanism and drug effects.
