摘要
人参皂甙Rg组、Rb组、Rg_1、Rb_1对大鼠肾脏微粒体Na^+,K^+—ATP酶均有抑制作用,IC_(50)分别为402,74μg/ml,15和50μmol/L。Rb组、Rg_1和Rb_1的抑酶作用可逆;增加K^+可竞争性拮抗Rg_1和Rb_1的抑酶作用,但Na^+对此作用无明显影响。动力学研究表明,Rg_1和Rb_1抑制Na^+,K^+—ATP酶,对底物ATP似为反竞争性抑制剂。
The results of present study showed that ginsenosides Rg,Rb,Rg,and Rb,all inhibited rat kindey microsome Na+,K+-ATPase activity in vitro and their IC50 were 402,74μ/ml,15 and 50μmol/L respectively.Ginsenosides Rb,Rg1 and Rb1 were reversible inhibitors on Na+,K+-ATPase.K+ could antagonize the inhibitory effect of Rg1 or Rb1 but Na+ showed no effect on the inhibition.Kinetic analysis revealed that both Rg1 and Rb1 are uncompetitive inhibitors with respect to ATP and competitive inhibitors with respect to K+.
