小檗碱对糖尿病肾病大鼠肾小球系膜细胞增殖和分泌TGF-β_1的影响 被引量:4

Effect of berberine on proliferation and secretion of transforming growth factor-β_1 of glomerular mesangial cells from diabetic nephropathy rats

摘要 目的探讨小檗碱对糖尿病肾病大鼠肾小球系膜细胞增殖和分泌TGF-β1的影响。方法分离培养糖尿病肾病大鼠肾小球系膜细胞,用MTT法检测系膜细胞的增殖能力,ELISA法测定系膜细胞分泌的TGF-β1,放射免疫法测定系膜细胞产生cAMP水平。结果间接免疫荧光鉴定结果显示所培养的细胞为系膜细胞。小檗碱(15、30、60、120μmol/L)能有效的抑制糖尿病肾病大鼠系膜细胞的增殖和TGF-β1的分泌,并显著升高糖尿病肾病大鼠系膜细胞的cAMP水平。结论升高糖尿病肾病大鼠系膜细胞cAMP水平可能是小檗碱发挥抑制系膜细胞过度增殖和分泌的机制之一。 AIM To study the effects of berberine on proliferation and secretion of transforming growth factor- β1 ( TGF-131 ) of glomerular mesangial cells from diabetic nephropathy (DN) rats. METHODS The glomerular mesangial cells from DN rats were collected and incubated. Mesangial cells proliferation was determined by 3- (4,5- dimethyhhiazal-2yl)-2,5-diphenyhetrazolium bromide (MTT) assay. TGF-β1 was examined by ELISA. Level of cAMP was determined by radioimmunoassay (RIA). RESULTS The results of immunofluorescence for vimentin and α-SMA were positive under fluorescence microscopy. Berberine (15, 30, 60, 120 μmol/L) significanfly inhibited the proliferation and secretion of TGF-βof mesangial cells from DN rats, and increased the level of cAMP. CONCLUSION Berberine could effectively ameliorate proliferation and secretion of glomerular mesangial cells from the DN rats. The mechanism of berberine for inhibiting the excessive expression of mesangial ceils from DN rat may be related to the effect of raising the level of cAMP.
