摘要
本研究确定了质粒pSET152从大肠杆菌(Escherichia coli)ET12567(pUZ8002)转移到链霉菌769中的接合转移的条件。在MS培养基上得到了最高的接合转移效率;孢子在50℃下热激10 min,供体菌和受体菌的比例为1:10,16~18 h后进行抗生素覆盖的情况下结合转移效率为1.03×10-6~1.23×10-5。经过连续5代的选择压力和PCR验证,表明质粒pSET152已经整合到受体菌株的染色体中。本研究首次建立了链霉菌769接合转移系统。
In this study,conditions for the conjugal transfer of pSET152 from E.coli ET12567(pUZ8002) to Streptomyces ahygroscopicus gongzhulingensis were established.The highest frequency was obtained on MS medium.The frequency of conjugant formation was 1.03×10-6~1.23×10-5 depending on heat treatment of the spores at 50 ℃ for 10 min,donor and recipient cells mixed in a 1:10 ratio,incubated 16 to 18 hours and then covered with antibiotics.After five generations selection under stress conditions,it was demonstrated by PCR experiments that plasmid pSET152 has integrated into chromosomes of the recipient strains.And this is the first report on the transfer system of S.gongzhulingensis.
