摘要
利用快速蛋白质液相色谱技术 (简称 FPL C技术 )对 3个典型黄萎病菌系 T9、VD8和 SS-4的外泌毒蛋白进行层析分离 ,皆得到两个致萎蛋白峰 :峰 致萎力弱 ,棉苗受害症状属黄斑型 ;峰 致萎力极强 ,症状属青枯型。 3个菌系强致萎峰 毒蛋白的百分含量由高到低依次为 T9、VD8、SS-4。对各菌系致萎峰蛋白做氨基酸组分测定 ,结果显示 :13个菌系的致萎峰 毒蛋白中的碱性组氨酸百分含量很高 ,平均达 14 .76% ,远大于峰 毒蛋白中的组氨酸含量 ,其平均值仅 3 .74 % ;2 3个菌系各致萎峰皆测不出甲硫氨酸 ,酪氨酸仅在 T9的峰 中含 0 .13 % ,其余各峰含量均为零 ,各峰的半胱氨酸含量很低 ,都在 1%以下。还利用高效液相色谱技术估测致萎峰蛋白的分子量 ,并探讨了黄萎病菌致萎毒蛋白的有关结构功能特征。
Toxic proteins secreted by three typical Verticillium dahliae strains T9,VD8 and SS 4 were separated by the technology of fast protein liquid chromatography(FPLC)to produce two wilting peaks.The first peak had a weak wilting ability to show the symptom of yellowish wilt on cotton seedlings with normal root system,the second one resulted in strong wilting ability to cause severe greenish wilt.However,the content of the second wilting peaks proteins followed as T9>VD8>SS 4.Analysis of amino component indicated that the second peaks toxic proteins included very high histidine to reach the average 14 7%,with the first ones only 3 74%.Additionally,tyrosine,only existed in the second peak of T9 with 0 13% content No methionine could be detected in all peak toxic proteins from these three strains,and cysteine were all under 1%.Lastly,the molecular weight of all peakstoxic proteins were estimated by the technology of high performance liquid chromatograph,and some properties of the construction and function of wilting proteins were also discussed.
