拟南芥中ELO家族同源基因的克隆与表达 被引量:1

Cloning Elongation(ELO) Homologue Genes in Arabidopsis thaliana and Construction of Expression Vectors Fused with eGFP

摘要 采用RT-PCR方法,克隆了拟南芥中可能参与超长链脂肪酸合成的ELO家族同源基因At1g75000、At4g36830和At3g06470的全长cDNA,构建其与eGFP基因融合的植物表达载体At1g75000∷eGFP∷NOS/pPZP211、At4g36830∷eGFP∷NOS/pPZP211和At1g06470∷eGFP∷NOS/pPZP211,并利用基因枪介导法将构建的植物表达载体转入洋葱表皮细胞,并在荧光显微镜下检测到了绿色荧光信号。为进一步研究拟南芥中AtELO家族同源基因的功能奠定基础。 Abstract:In order to investigate whether ELO homologue genes of Arabidopsis thaliana are involved in very long chain fatty acid biosynthesis, the cDNA sequences of Atlg75000,At4936830 and At3g06470 were amplified by RT-PCR. We made constructs fused with eGFP in plant transient expression vectors Atlg75000 eGFP 1 : NOS/pPZP211, At4g36830 : eGFP : NOS/pPZP211 and Atlg06470 eGFP NOS/ pPZP211 respectively,transformed above vectors in onion epidermal cells by gene gun and examined green fluorescence.
