摘要
以海棠花组培苗和叶片为材料,应用正交试验法,以MS培养基为基础,分别加入不同浓度配比的6-BA/IBA组合和TDZ/IBA组合作为增殖培养基和叶盘不定芽再生培养基,研究了不同植物激素及其配比对组培苗增殖、叶片不定芽再生的影响。结果表明:在添加6-BA 1.0mg.L-1+IBA 0.1mg.L-1的MS培养基上最适宜海棠花组培苗的增殖及生长,增殖倍数高达8.11,海棠花组培苗平均高度达2.14cm;在添加TDZ 1.0mg.L-1+IBA 0.5mg.L-1的MS培养基上叶再生效率最高,再生效率为100%,每叶平均再生芽数为6.73。
Influences of different plant growth regulators on shoot proliferation and adventitious shoot regeneration of Malus spectabilis were investigated. The shoots and leaves were used as explants in the orthogonal experiment. The culture medium was MS supplemented with different concentrations of 6-BA, TDZ and IBA. The results showed that MS adding with 6-BA 2.0 mg·L-1 + IBA 0.1 mg· L-1 was the optimum medium for shoot proliferation, the multiply of proliferation value reached 8. 11, and the average height of shoots was 2.14 cm; The highest percentage of regenerated shoots (100%) and the highest number of shoots per explant (6.73) occurred on MS medium containing 1.0 mg ·L 1 TDZ and 0.5 mg · L 1 IBA.
